Intestinal microbe-mucosa crosstalk in health and disease

Abstract:
A tightly regulated balance between the intestinal epithelium, immune cells and the microbiota maintains intestinal homeostasis. Disruption of this balance associates with diseases like inflammatory bowel disease (IBD), which is characterized by epithelial barrier impairment and aberrant inflammatory responses to microbiota. The regulation of innate immune sensing of commensal bacteria in the intestinal mucosa is key to prevent this, and relies on the maitenance and repair of epithelial barrier function. I will discuss how innate immune responses to bacterial exposure in the context of acute bacterial infection and intestinal inflammation shape epithelial phenotypes and induce repair programs. I will then put into perspective how these epithelial repair programs might affect microbe-mucosa crosstalk, and how recent developments in organoid tools can be exploited to dissect this crosstalk mechanistically.
